Key words in English:
rapeseed; abscisic acid; microspore embryos; fatty acid; gas chromatography
Annotation in original language:
The content of the fatty acids was determined using gas chromatography in the rapeseed (Brassica napus) cotyledons of microspore embryos. Impacts of abscisic acid application and different dates of sampling on the individual fatty acids content in embryo cotyledons were evaluated. The effect of different concentrations of abscisic acid (7.5 and 15 mu M) on the fatty acid composition was evaluated. At the selected genotypes the content variability of main fatty acids was detected. It was found that treating with abscisic acid caused the increase of stearic acid in comparison to samples where abscisic acid was not applied. Simultaneously, the decrease of the concentration palmitic, oleic linolenic, and linoleic acid was evaluated. The achieved results confirmed the possibility of analyzing the content of the fatty acids in the microspores embryos using gas chromatography and applying this method to predict the content of the fatty acids of the dihaploid material for practical breeding of rapeseed.
