The microbial assemblies on the surface of plants correlate to specific climatic features, suggesting a link between environmental conditions and microbial inhabitation patterns. On the other hand, microbial communities demonstrate distinct profiles depending on the plant species and regional origin. In order to investigate the impact of biogeography on yeast diversity we analyzed the spreading of different yeast species on the surface of fruits and berries cultured in distinct geographical regions as Lithuania and Czech Republic. For primary detection and enumeration of microbial flora present on plant surfaces, traditional microbiological and microscopy methods were used. For research on microbiota, molecular biology techniques were employed, namely: extraction of genomic DNA, amplification of ribosomal DNA genes and RFLP analysis, metagenomics identification of microorganisms with subsequent phylogenetic analysis. During this study we identified eukaryotic microorganisms common for many plants, as well as yeast inhabiting the surface of particular fruits and berries only. It was distinguished the prevalent yeast present on apples, plumps, pears, currants, buckhorns, etc, capable survive the fermentation conditions. Comparison of yeast present on two plants of the same type but located in different geographical regions and climatic conditions (Lithuanian and Czech Republic) allowed investigation of the impact of climate change to microbiota composition of fruit-berry ecosystem.
