Nowadays, HPLC method is apply in many field of application as medicine, chemistry, environment etc. The use of this method for quantitative determination photosynthetic pigment contents is one of the possibilities. There is a lot of methods, that are different in usage of column, chemical composition of solvent A and B, flow rate, detection wavelength or the type of pump (isocratic, gradient). In addition, in literature are not mentioned importance parameters, which affected results. Consequently, in this work, we are concerned with influence of these parameters: temperature, setting of spectral bandwidth of detector, flow rate and composition of solvent B. The HPLC pigment analysis was carried out on the basis of Färber and Jahns (1998), which was modified ourselves as follow: the gradient was adjusted from 10 to 12 minutes instead of gradient from 9 to 12,5 minutes. This set up is necessary to correctly integration of lutein and zeaxanthin.
