Carboxylase activity of enzym Ribulose-1,5-bisphosphat carboxylase/oxygenase (Rubisco, EC 4.1.1.39) has a key importance in photosynthesis. The method of spectrophotometrical determination of activity Rubisco was in 2003 established in laboratory of Deparment of Physic, Faculty of Science of University of Ostrava. From the pilot measurements of Rubisco activity of barley and spruce plants were made some changes of the Hrstka et al. (2002) protocol. The measurements were carried out on barley (Hordeum vulgare) and spruce (Picea abies) grown under laboratory conditions and on spruce (Picea abies) acclimated to elevated CO2 concentration (700 umol (CO2) mol-1). For determination of Rubisco activity was used spectrophotometr UV 550, program Vision 32. Program had adjustment for measuring of absorbance (methodology FIXED) and spectra (methodology SCAN).
